An international group of researchers, including those from Moffitt Cancer Center in Tampa, Fla., have published a paper in the March 14 issue of the New England Journal of Medicine reviewing the results of a study that analyzed mutations in 18 genes of 398 patients who had acute myeloid leukemia (AML). They found that several mutated genes predicted improved outcomes when patients with certain gene mutations were given high-dose induction chemotherapy. Their findings suggest that mutational profiling could potentially be used for both risk stratification and also in helping health care providers make therapeutic decisions for some AML patients.

“Previous studies have found that AML is a highly heterogenic disorder,” said study co-author Hugo F. Fernandez, a senior member at Moffitt and associate chief of Moffitt’s Blood and Marrow Transplantation Division. “Moreover, recent studies have revealed that a number of genetic mutations in AML patients might have prognostic value. The question of the presence of these gene mutations altering outcomes based on current therapy had not been answered to date.”

Their paper cites a clinical trial carried out by the Eastern Cooperative Oncology Group (ECOG) in which dose-intensified chemotherapy improved outcomes in two age sets of AML patients. Based on these findings, the research team hypothesized that carrying out mutational analysis of all known molecular alterations occurring in more than 5 percent of patients with AML might allow for the identification of distinct, molecularly defined subgroups of patients who might benefit from dose-intensified chemotherapy.

The laboratory research team subsequently performed a mutational analysis on diagnostic samples from 398 patients enrolled in the ECOG clinical trial they cited and used patients’ frozen sample cells for extraction and profiling. The researchers validated the results of this latter group of 104 patients.

“We found that intensification of the dose of anthracycline significantly improved outcomes and overall survival in patients with mutations in DNMT3A, NPM1 or MLL translocations,” said Fernandez. “This finding suggests that mutational profiling could be used to determine which AML patients will benefit from dose-intensive induction therapy.”

“Most importantly,” said Fernandez, “this study demonstrates how integrated mutational profiling of samples from a clinical trial cohort can advance understanding of the biologic characteristics of AML.”

(AP) ? When Phong Yang, a Hmong refugee from Laos, landed in California’s Central Valley ? via stops in Thailand and France ? he was 14 years old. He learned to speak Hmong from his parents, but today he has a hard time teaching the language to his children, who are distracted by cell phones and computers.

Many Hmong are losing their language, Yang said, leading to fears that their cultural identity will be lost.

A new technological tool may help bridge generation gaps and encourage preservation of their language among the Hmong. The community in Fresno, in partnership with researchers at Microsoft, has launched an online Hmong translator.

It’s a significant step, community leaders say, because the language that withstood assimilation efforts throughout history is now at risk of being lost. Hmong ? an ancient language, whose oral tradition goes back thousands of years ? developed a written form only 60 years ago.

“It tells our elders and our children, your language is important, it’s on the map now,” said Yang, a teacher of Hmong at California State University, Fresno who served as coordinator for the project. “And you can have fun while using it.”

The Central Valley has been home since the 1970′s to one of the largest communities of Hmong ? an Asian ethnic minority group ? who had fled their mountain villages in Laos and languished for years in Thai refugee camps. Approximately 33,000 Hmong live in Fresno, according to census figures, the second largest Hmong population in the country. A total of 260,000 Hmong reside in the United States.

The history of the Hmong has been marked by persecution. But while the Hmong were uprooted many times and do not have a country of their own, they never lost their language. Now, as thousands of Hmong have resettled in western countries, the language is facing new challenges.

“All these years, the language has been preserved, despite efforts to eradicate it,” said Will Lewis, a Microsoft program manager who worked on the Hmong translator. “Now, the irony is that in the United States, a country where they’re free to speak it, the thing that never happened in Hmong history is happening; some children are not learning Hmong.”

The translator uses a statistical model to find patterns and assign probability to words in context. Since November, when the project started, community members and researchers have fed a computer with hundreds of documents in the two languages, as well as with entries from an online Hmong-English dictionary. Since dictionaries offer no context, community members entered sentences for each word.

A similar concept could be applied to benefit other minority and indigenous languages, most of which aren’t covered by automatic translators such as Microsoft’s Bing Translator or Google Translate, Lewis said. Less than 100 languages ? mostly the dominant, widely-used ones ? are currently covered, out of the more than 7,000 languages world-wide.

The technology, said Hmong elder Chue Her, will help both older Hmong who don’t speak English and U.S. born youngsters who speak little Hmong. Smart phone apps that use the translator are now also available.

Using the Microsoft translator, Hmong elders can translate web sites, documents and even use the translator to communicate with children and grandchildren.

But the biggest benefit will be to Hmong youth, Yang said. At home ? where immigrant children were traditionally exposed to their parents’ language ? young people now spend their time in front of computers.

“When the kids are at home, instead of spending time with their parents and speaking in Hmong, they use the Internet and talk to their friends on Facebook. And it’s all in English,” Yang said.

Because written Hmong is new, there are few Hmong textbooks. The hope, Yang said, is that the translator will fill the void and make the language interesting and cool to learn.

“The translator makes it more fun than just giving the children books. Now parents can tell them, look, you can still socialize with your friends, but you can do it in Hmong,” Yang said.

And the language can support more than just communication, community members said.

“Along with learning the language, the children get immersed in the culture and that promotes their identity,” said Vicky Xiong, a second grade teacher in the Clovis Unified School District. “The language will give them pride in themselves and their people.”

Xiong said she realized the importance of teaching the language when her U.S. born 6th grader son ? now a freshman at Fresno State ? told her, “Mom, I don’t want to be Hmong anymore. Let’s speak only in English.” She enrolled him in a language class offered by Stone Soup, a Fresno Hmong nonprofit, but few such classes are available, she said.

Joshua Lor, Xiong’s son, said he eventually realized the importance of the language.

“My grandpa told me stories about the Hmong, about how he served in the war, and how they moved from Laos to Thailand to America,” Lor said. “The language opened my eyes to the history of Hmong culture. It’s exciting that the translator can help kids do that.”

Vatican: Pope will bond with Mexico on first trip

LEON, Mexico (AP) ? Pope Benedict XVI headed off on his first trip to Mexico on Friday, using a cane as he boarded the plane for a trip that a representative says will help him build his own bonds with a nation that had a passionate adoration of his predecessor.

Pope John Paul II was greeted with ecstatic scenes on each of his five trips to Mexico, and love for the late pontiff was so strong that millions turned out to honor him even after death when a glass case containing his blood was brought to the country.

He was a vigorous 58-year-old when he first arrived. Benedict is now 85, his age emphasized by the black cane he carried as he set out on his own pilgrimage on Friday.

Papal aides, speaking on condition of anonymity, said the pope started using the cane about two months ago in private because it makes him feel more secure, and not for any medical problem.

So far Mexicans have shown restrained excitement about the pope’s arrival in Guanajuato, a deeply conservative state in sun-baked central Mexico. Dedicated campgrounds with a capacity for hundreds of thousands of pilgrims were virtually empty.

Carlos Aguiar, president of the Mexican Episcopal Conference, said he expected the faithful to begin arriving Friday.

The papal envoy, Christophe Pierre, said Benedict’s visit would allow Mexicans to “discover the pope” and “his ability to communicate, to speak deeply, but simply.”

There were some stirrings of enthusiasm about Benedict’s first trip to Mexico as locals tidied up the city in preparation. About 60 people burned brush to create a neater vista along a section of the pope’s route from the airport to the Catholic school where he will spend his three nights in Mexico.

Maria Belen, a 36-year-old homemaker, swept inches of powdery dust and gravel off a sidewalk.

“It’s the first time a pope has visited our community. We’re excited,”

Farther down the highway, young men and women sold flags in the yellow-and-white Vatican colors, although few motorists stopped to buy them.

The biggest crowd appeared to be the thousands of youths attending a convention of evangelical Protestant missionaries a the highway exit leading to Bicentennial Park, where Benedict will deliver Mass on Sunday.

Light of the World Church spokesman Ezequiel Zamora said the convention had been planned long before the pope’s visit was announced, and the evangelical church had no intention of trying to upstage the pontiff.

Guanajuato is 93.8 percent Catholic, the highest percentage in Mexico, but Protestant and evangelical denominations have made deeper inroads in other parts of the country, particularly along the northern and southern borders.

The last time a pope visited Mexico, more than 1 million believers cheered and wept in the streets of Mexico City. Dancers dressed in Aztec costumes shook rattles and blew conch shells inside the cathedral where John Paul II canonized the first Indian saint in the Americas.

A decade later, Benedict comes to a church battling to overcome painful setbacks that include legalized abortion and gay marriage in the capital of the most populous Catholic country in the Spanish-speaking world.

It is also a nation grappling with a drug war that has spread fear into once-tranquil regions such as Guanajuato.

“There is a very immense peace that we need in Mexico because of the insecurity,” said Marcela Arguello, a 26-year-old housewife who said she planned to stand along the route of the papal motorcade through the city of Leon, the state’s largest city.

Mexico has been traumatized by the deaths of more than 47,000 people in drug-related violence in less than six years, and while Guanajuato is far from major drug trafficking routes, the shadow of the conflict looms even here.

“Yes, he’ll talk about violence, we can’t ignore that, but I can tell you as a representative of the Holy Father that there’s much more than violence in Mexico,” Pierre said.

“The Holy Father, in the name of God, comes to remind, to ask people not to lose their path in life,” Pierre added.

As many as 300,000 people are expected to gather for the Sunday Mass.

Guanajuato’s constitution declares that life begins at conception and bars abortion with extremely limited exceptions. Seven women were jailed there in 2010 for the deaths of their newborns and later released. The women said they had miscarriages, not abortions.

Benedict’s church is encouraging more such laws across Mexico, reacting partly to the legalization of gay marriage and abortion in Mexico City, the cultural and political center of the country.

At the same time, church leaders are fighting to overcome a scandal over the most influential Mexican figure in the church.

The Rev. Marcial Maciel founded the Legionaries of Christ order, which John Paul II praised as a model of rectitude. But a series of investigations forced the order to acknowledge in 2010 that Maciel had sexually abused seminarians and fathered three children. Church documents released in a book this week reveal the Vatican had been told of Maciel’s drug abuse and pederasty decades ago.

President Felipe Calderon’s government is backing legislation that would end legal restrictions on religious observances in public places, as well as a ban on religious participation in politics.

If approved, it could lay the groundwork for laws allowing church ownership of media and openly religious education on school property, said political analyst John Ackerman at the legal research institute at Mexico’s National Autonomous University.

“This opens the door for the church to start using public spaces,” Ackerman said. “It has the full intention to be interpreted as occupying public spaces with religious ceremonies, that’s what’s on the table.”

The hacker group Anonymous in Mexico crashed at least two of the websites for Benedict’s visit to Mexico on Thursday, claiming his trip is meant to support Calderon’s conservative National Action Party, which is especially strong in Guanajuato, at the start of a presidential election campaign.

Anonymous Mexico also said in a video posted on social media sites that the pope’s visit will cost Mexicans money that could be better spent on the poor.

The Major League Gaming Pro Circuit Winter Championships start Friday and run throughout the weekend. Taking place in Columbus, Ohio, the world’s finest “StarCraft 2,” “Halo: Reach,” “Mortal Kombat,” “Soulcalibur V” and “King of Fighter XIII” players have all converged to see who is the best professional gamer in their respective circles.

To reach this point, every player was a victor in various semi-final contests. In the case of the “StarCraft 2″ players, all the participants were chosen as the result of the Winter Arena Feb. 24-26 at Major League Gaming’s headquarters in New York City. MSNBC was on hand to view the event and to speak with those involved.

Advocates of the e-sports movement have long argued that it’s on the same level as traditional sports, comparable to baseball, football and the rest. While many will concede that it’s somewhat silly to compare someone who excels on the basketball court with someone who excels at “Street Fighter,” becoming a serious competitor requires much hard work and dedication. And thus an appropriate amount of respect.

The challenges that athletes on both sides face are actually quite similar, including their parents. “It was a constant concern with mine, when I first became involved,” said Andrew Dudynsky, a former professional “Halo” player, who has since retired and is currently pursing various career paths, all still related to e-sports, including commentating (which he’ll be handing for “Reach” this weekend’s event) .

Dudynsky’s mother gave him a choice, he said: either continue professional gaming and go to community college, or stop playing entirely to attend a major school. He was confident that he’d be able to dedicate himself to both serious gaming and a major university, and eventually did so. His average GPA was 3.97.

One curious consistent trait among all the participants?interviewed is how every single one proudly stressed the high marks they were able to achieve in school while competing. An understandable point of pride, given all the hard work and discipline, even when one is “simply playing a game.”

Like many professions, there’s little glory at first. Dudynsky described all the time and money he spent while trying to establish himself early on. It’s especially difficult to maintain a personal life, like friends and girlfriends, because of the grueling schedule.

During the height of his pro gaming career, he said he would spend every waking moment that wasn’t dedicated to studies to playing. Or traveling to whatever part of the world a competition was taking place. Often he’d take a red eye back, to arrive just in time for his first class in the morning. But his perseverance paid off.?

While he didn’t say specifically how much money he made during his heyday, he said, with confidence in his voice, that it was a good amount, enough to pay for college at the very least.

These days, Dudynsky is no longer an active competitor, but instead coaches for the current crop of players. He also explained how, in addition to the hefty cash prizes that contests can provide, a good deal of income stems from sponsorships. “Professional gaming is still young enough in which advertising for someone still matters.”

He elaborated: ?If a winning gamer is seen wearing the logo of a particular kind of video card or keyboard and mouse, that company will see an immediate return. But if Tiger Woods is seen with a particular kind of golf club, the effect is not nearly as strong.?

Another form of income?is live-streams, he said. Almost all are found on Twitch.tv, a relatively recent offshoot of Justin.tv. Not only does it broadcast all of Major League Gaming’s programming, but it’s where anyone who is anyone in the scene has a presence. Not only do pro gamers share strategies and commentary, but it helps to get them better-known, which enhances the brands they represent even further.

Justin Kan, co-founder of Justin.tv, explains: “We consider it like ESPN for video games, except for having ESPN 1, 2, and Classic, we have Twitch.tv 1, 2, 3, 4, all the way to 700.” Programming ranges from content provided by the sponsors themselves, to just candid, fly-on-the-wall looks at the lives of pro gamers, many of whom live and train together under one roof.

Some of these personalities bring in about “$5,000 to $7,000 a month, from just streaming their game playing,” Kan said. “Some earn high five figures ? I think we might have one or two who are bringing in six figures on the site,”?just from people watching them play “StarCraft” or “Halo.” Kan believes such numbers are encouraging the business.

New iPad’s heat issue caught on thermal camera

According to tests conducted by Consumer Reports, the new iPad runs hotter than its?predecessor. A recently released thermal photo shows the heat difference quite clearly.

As you can see in the image above, the new iPad hits temperatures of about 116 degrees Fahrenheit after about 45 minutes of running an intense-action game. This is up to 14 degrees F warmer than the iPad 2 will run under similar conditions, based on Consumer Reports’ analysis.

“During our tests, I held the new iPad in my hands. When it was at its hottest, it felt very warm but not especially uncomfortable if held for a brief period,” Consumer Reports reviewer Donna Tapellini explained according to Reuters.

An Apple spokeswoman told Reuters that the new iPad runs “within [Apple's] thermal specifications.” The Apple website lists?the third-generation iPad’s operating temperature as 32 to 95 degrees F.
